چکیده

Aims and Objectives: The effectiveness of crude methanolic extracts of Bridelia ferruguinea, Tridax procumbens, Ocimum gratissimum and Lawsonia inermis against clastogenicity induced by sodium arsenite (SA) was evaluated using an in-vivo rat bone marrow cell micronuclei test and the DPPH radical scavenging assay. Methods: 50 mg/kg body weight of the extracts were given by gavaging to the rats in five groups for seven consecutive days as a dietary supplement followed by a single dose of SA (2.5 mg/kg per body weight) which was administered intraperitoneally. Results: Preliminary screening of the clastogenicity showed that aqueous extracts of these plants have no significant clastogenic activity in rat. Pre-treatment of rats for seven days with extracts orally before exposure to SA resulted in a significant reduction of the degree of formation of micronuclei in polychromatic erythrocytes of the bone marrow. The degree of reduction of arsenite was of the order B. ferrugenia > O. gratissimum>T. procumbens > L. inermis. Phytochemical analysis carried out on the extracts revealed the presence of known chemical constituents. The phytochemical constituents might be responsible for the strong DPPH scavenging activities exhibited by the plant extract. Conclusion: This study suggests that B. ferruguinea, T. procumbens, O. gratissimum, and L. inermis extracts have anticlastogenic potential and possibly due to their antioxidant properties. Therefore, the plant extracts may be useful in the stoppage of arsenite-induced toxicity in areas where arsenic is a potential environmental contaminant.

Hepatoprotective and anticlastogenic effects of ethanol extract of Irvingia gabonensis (IG) leaves in sodium arsenite-induced toxicity in male Wistar rats.

Consumption of arsenic contaminated water has been associated with diverse health defects such as cancer and skin lesions. Some plants of medicinal value have been reported to show protective effects against toxins. In this study, the effects of ethanol extract of the leaves of Irvingia gabonensis (IG) against sodium arsenite (SA) induced hepatotoxicity and clastogenicity in male Wistar rats wa...

The effects of Vitamin C on quality of semen, testicular tissue, and level of apoptosis in mice exposed to sodium arsenite

Background: Arsenic as a heavy metal exerts malformation and changes in the reproductive system of men via induction of oxidative stress. We sought to investigate the impact of Vitamin C (Vit C), as a potent antioxidant, on sperm quality and testicular tissue in mice that received sodium arsenite. Methods and materials: NMRI mice were classified into four groups: control, sodium arsenite (7 mg...

Arsenic and Oxidative Stress in Pentylenetetrazole-induced Seizures in Mice

Background and Objective: Chronic arsenic toxicity is a widespread problem; the role of brain oxidative stress has been suggested in the genesis of epilepsy and in the post-seizure neuronal death. However, studies investigating the effects of arsenic on seizure and related mechanisms are limited. The purpose of this study was to examine the effect of prolonged exposure to sodium arsenite on oxi...

Exposure of Laboratory Mice to Domestic Cooking Gas: - Implications for Toxicity

The ability of domestic cooking gas to induce hepatotoxicity and clastogenicity in mice was studied. The mice were exposed to domestic gas for twenty-one days at doses of 100 mg/kg, 200 mg/kg and 300 mg/kg respectively. The positive control group of mice were given sodium arsenite intraperitoneously at a dose of 2.5 mg/kg body weight. While the negative control group had only distilled water, s...
